の can be either possessive (Aerith's...) or used to modify or clarify other nouns.

は is marking the topic of the sentence. (This is actually a pretty difficult thing to explain, so I won't bother )

５年前に消息を絶った
...went missing 5 years ago...

消息を絶つ is kind of a set-phrase meaning that you lose contact with someone and they go missing and you don't know what's going on with them.

ソルジャー・クラス1stの青年ザックスだ。
SOLDIER Class 1st youth Zack [is].

In Japanese, you'll see this kind of thing a lot. Descriptions will come before what they're describing. 「５年前に消息を絶ったソルジャー・クラス1stの青年」 was all describing 「ザックス」.
